防己对培养的绿色荧光蛋白小鼠胸腺细胞凋亡的影响

【摘要】 目的:研究防己对绿色荧光蛋白(GFP)小鼠胸腺细胞凋亡的影响。方法:采用原代培养技术、荧光显微镜、DNA凝胶电泳以及免疫细胞化学方法观察不同浓度防己对GFP小鼠胸腺细胞凋亡程度的影响。结果:防己在1-100μg/ml可不同程度抑制小鼠胸腺细胞的增殖,荧光染色可见典型凋亡小体, DNA凝胶电泳有典型的DNA梯形条带,免疫细胞化学可观察到Fas与FasL阳性表达细胞。结论:不同浓度防己(1-100μg/ml)可不同程度抑制GFP小鼠胸腺细胞的增殖,促进其凋亡,其凋亡可能通过Fas/FasL系统而诱导。

【Abstract】 Objective: To investigate the effect of Stephania Tetrandra S. Moore (S Tet, Han-Fan-Chi) decoction on GFP mice thymic cells. Methods: The inhibition of thymic cell proliferation was detected by means of fluorescence microscope, DNA gel electrophoresis and immunocytochemistry. Results: S Tet decoction inhibited thymic cell proliferation in a dose - and time-dependent manner. Evident apoptotic changes (typical apoptotic bodies and DNA ladder) was observed; Fas and Fas-L positive cells were also be observed. Conclusion: S Tet decoction at different doses range from 1-100μg/ml can significantly inhibit the proliferation and induce the apopto-sis of thymic cells in mice, which is correlated with Fas/Fas-L system.
