那曲地区藏族mtDNA D-环区高变区Ⅰ序列多态性
Sequence polymorphisms of mitochondrial DNA D-loop region hypervariable region Ⅰ in Naqu Tibetan nationality in China
【摘要】 目的:研究藏族人群线粒体DNA D-环区序列遗传多态性。方法:应用PCR扩增产物直接测序法,对36名藏族无关个体线粒体DNA D-环区高变区Ⅰ(hypervariable regionⅠ,HVR-Ⅰ)进行测序分析。结果:HVR-Ⅰ中404个核苷酸序列与Anderson相应序列比较共发现有51处突变,构成34种单倍型。藏族线粒体DNA的HVR-Ⅰ基因差异度为0.996 1,偶合概率为0.031 6。结论:线粒体DNA D-环区HVR-Ⅰ序列的多态性在人类学、法医学领域具有较高的应用价值。
【Abstract】 Objective: To study the genetic polymorphisms of the mitochondrial DNA(mtDNA) D-loop region in Naqu Tibetan nationality.Methods: Sequence polymorphisms of 36 unrelated Tibetan mtDNA D-loop hypervariable regionsⅠ(HVR-I) were determined by PCR and direct sequencing.Results:Fifty-one polymorphic sites and 34 haplotypes were identified in the sequences of 404 nucleotides for HVR-Ⅰ.The genetic diversity was calculated as 0.996()1,and the genetic identity was 0.031()6.Conclusion: Sequence polymorphisms of mtDNA D-loop region HVR-Ⅰare very useful in anthropology and forensic practice.
