重组Tgo DNA聚合酶的特性

【摘要】 检测了重组Tgo酶的扩增性能、耐热性以及长距离PCR能力,结果表明,该重组酶扩增性能与Taq酶相当;其耐热性极强,在95℃40 m in,仍具有很强的聚合活性;利用重组酶在常规PCR条件下,成功扩增了5.3 kb的拟南芥Tim eless基因片段.

【Abstract】 The amplification capability,the heat tolerance and the ability to long PCR of Tgo DNA polymerase were examined.The results reveal that Tgo DNA polymerase has the same amplification function as Taq(enzyme) but with a high fidelity.It also has a strong resistance to heat.Its amplification function would not be notably affected at 95 ℃ for 40 min.Via the recombinant enzyme, the(5.3 kb) segment of Timeless gene was successfully amplified from Arabidopsis under the general conditions of PCR.
