芦荟粗多糖对体外培养人表皮细胞增殖活性及细胞周期的影响

【摘要】 目的研究芦荟粗多糖对体外培养人表皮细胞增殖活性及细胞周期的影响。方法在芦荟粗多糖(75、150、300、600、1200 μg/ml)的作用下,光镜观察表皮细胞形态及细胞融合时间,电镜观察细胞超微结构,MTT法以及[3H]-TdR渗入量观察细胞增殖状况,流式细胞仪检测细胞周期。结果与对照组比较,芦荟粗多糖作用后表皮细胞形态学变化不明显,但芦荟粗多糖组细胞融合时间明显缩短(P< 0.05);细胞超微结构观察发现,300、1200 μg/ml芦荟粗多糖组的细胞核内以常染色质为主,而对照组与 75μg/ml芦荟粗多糖组则以异染色质为主;从生长曲线上看,芦荟粗多糖作用2 d后细胞增殖明显增快; [3H]-TdR渗入量与对照组比较呈显著性上升(P<0.05);流式细胞分析显示,G0/G1期细胞所占百分率显著性减少,G2/M期和S期细胞所占百分率显著性增加(P<0.01)。结论芦荟粗多糖可通过影响表皮细胞的DNA合成及细胞周期来促进细胞增殖。

【Abstract】 Objective To investigate the effects of Aloe coarse polysaccharide on proliferation and cell cycle of keratinocytes in vitro. Methods After using 75, 150, 300, 600, 1200 μg/ml Aloe coarse polysaccharide, the morphology and the confluence time of keratinocytes were examined under light microscope, and the ultrastructures of keratinocytes were studied under electron microscope. The proliferation of keratinocytes was measured by MTT analysis and [3H]-TdR infiltration and the cell cycle of keratinocytes was analyazed by flow cytometer. Results Compared with control group, there was no significant difference in the morphology of keratinocytes, and the time of keratinocytes confluent was significantly shortened in Aloe coarse polysaccharide group. In the 300 and 1200 μg/ml Aloe coarse polysaccharide groups, the main components in keratinocytes nucleus were euchromatin, but were heteroehromatin in control group and 75 (μg/ml group. The proliferation curve showed that the keratinocytes grew markedly at 2 days of incubation. The [3H]-TdR infiltration levels were significantly increased in Aloe coarse polysaccharide groups than those in control group ( P < 0.05) . The flow cytometry analysis of the cell cycle distribution also revealed that the percentage of phase G0/G1 was markedly decreased, and G2/M and S was significantly increased ( P < 0.01) in Aloe coarse polysaccharid groups. Conclusion Aloe coarse polysaccharide could promote the proliferation of keratinocytes by DNA synthesis and the cell cycle.
