恒磁场对脂多糖诱导内皮细胞与中性粒细胞黏附及细胞间黏附分子表达影响的研究
Effect of static magnetics on adhesion of endothelium and neutrophil and expression of intercellular adhesion molecule-1 with LPS
【摘要】 目的研究恒磁场对脂多糖(LPS)诱导中性粒细胞与内皮细胞黏附及内皮细胞表达细胞间黏附分子的影响。方法人脐静脉内皮细胞于0·05、0·1、1mT的磁场中曝磁,用LPS刺激内皮细胞,内皮细胞与中性粒细胞黏附用细胞计数法评估,内皮细胞表面细胞间黏附分子表达用流式细胞仪及酶联免疫吸附法测定。结果LPS刺激后,中性粒细胞的黏附率为58%;0·05、0·1mT磁场条件下中性粒细胞黏附率下降为40%及38%,与单纯LPS组比较明显下降(P<0·05),而1mT组中性粒细胞黏附率为65%,与单纯LPS组相近。单纯LPS组细胞间黏附分子表达为10·34±0·33/0·89±0·16;0·05、0·1mT磁场条件下细胞黏附分子表达为6·61±0·17/0·53±0·18、6·98±0·15/0·46±0·10,与单纯LPS组比较明显下降(P<0·05);1mT组为10·99±0·41/0·78±0·15,与单纯LPS组比较无明显差异。结论0·05、0·1mT可以抑制LPS刺激下内皮细胞与中性粒细胞的黏附及内皮细胞表面细胞间黏附分子的表达。
【Abstract】 Objective To investigate the effects of static magnetics on adhesion of neutrophil and endothelium and expression of intercellular adhesion molecule-1(ICAM-1) on human umbilical vein endothelial cell(HUVEC). Methods HUVEC were exposed to 0.05mT, 0.1mT, 1mT static magnetics adhesion of neutrophil was calculated after HUVEC was stimulated by LPS. Flow cytometry and ELSIA were used to study expression of ICAM-1 on endothelium with stimulating of LPS. Results The adhesion of neutrophil l on HUVEC was attenuated by 0.05mT, 0.1mT static magnetics(58% vs. 40%, 38%, P<0.05). Furthermore the expression of ICAM-1 was decreased by 0.05mT, 0.1mT static magnetics. No change was found when HUVEC exposed in 1mT static magnetic field compared with control group. Conclusion 0.05mT, 0.1mT static magnetics can attenuate adhesion of mononuclear cell and endothelium and expression of vascular cell adhesion molecule-1(ICAM-1) on HUVEC. While 1mT doesn′t influence neutrophil adhesion and the expression of ICAM-1.
