人胚肺成纤维细胞突变株Z-HL16C转化性质的初步鉴定
Preliminary identification of the mutant human embryonic lung fibroblast Z-HL16C cell strain
【摘要】 目的初步鉴定人胚肺成纤维细胞(HL)突变株Z-HL16C的转化性质。方法采用细胞形态学观察、细胞遗传学染色体分析、细胞在裸鼠体内恶性生长情况的组织病理学检测以及细胞对病毒的敏感性等方法。结果HL细胞转化的Z-HL16C细胞在形态上由梭形的纤维细胞变成多边形的上皮样细胞,且具有稳定无限传代等特点;染色体数目由正常人的46条二倍体,转化成多数为亚三倍体或超三倍体;染色体结构核型中约有30%~40%的染色体为结构异常染色体;裸鼠移植肿瘤发病率为100%,其瘤组织病理检测结果显示,该细胞株为癌变细胞株。病毒实验表明,Z-HL16C细胞对单纯疱疹等多种常见病毒高度敏感。结论人胚肺成纤维细胞的突变株Z-HL16C是对病毒高度敏感的癌细胞株。
【Abstract】 Objective To study the characteristics of Z-HL16C cell strain mutated from the human embryonic lung fibroblasts(HL).Methods Morphology observation,chromosome cytogenetics,tumorigenesis in nude mice,and the sensitivity to virus were examined.Results As a transformed cell strain,the Z-HL16C showed polygon shapes liking epithelium;the number of chromosome was sub-triploid or hyper-triploid,and the structures of 30%~40% chromosomes were abnormal.The incidence of its inoculation in nude mice was 100%,and the pathological studies proved its tumorigenesis.The cytopathic effects of many kinds of viruses were observed in Z-HL16C cell strain.Conclusion The Z-HL16C is one of the cancer cell strains transformed from HL cells.
