D-半乳糖致蛋白质糖基化动物模型的建立

【摘要】 目的建立大鼠蛋白糖基化模型。方法给SD大鼠腹腔注射D-半乳糖150 mg/kg 8周。结果模型组动物2 h血糖、晶体醛糖还原酶活性、血清果糖胺水平、糖化血红蛋白及晚期糖基化终末产物含量均明显高于正常对照组(P<0.05),而氨基胍处理具有明显的改善作用。结论大鼠连续腹腔注射D-半乳糖8周,导致糖基化主要产物明显增高,可以作为糖基化动物模型用于实验研究。

【Abstract】 Objective To establish a new rat model of glycation.Methods D-galactose((150 mg/kg)) was established in rats by intraperitoneal injection for eight weeks.Methods After continuous administration of D-galactose for eight weeks,the level of two-hour blood glucose、activity of aldose reductase、level of fructosamine in serum、the amount of glycohaemoglobin and advanced glycation end-products are all higher than that of control(P<0.05).However,for these changes aminoguanidine has impressive improvement effect.Conclusion Continuous intraperitoneal administration of D-galactose for eight weeks can cause the amount of main products of glycation to increase significantly,so the rat model induced by D-galactose in can be used to study the process of glycation.
