甲基化特异性PCR法和限制性内切酶-PCR法检测脑胶质瘤hMLM1基因启动子区甲基化

【摘要】 目的研究脑胶质瘤中hMLM1基因启动子区的甲基化状态,并比较不同的甲基化检测方法。方法采用甲基化特异性PCR(methylation specific PCR,MSP)法和限制性内切酶-PCR法,检测42例脑胶质瘤和5例正常脑组织中hMLM1基因启动子区的甲基化状态。结果用MSP法和限制性内切酶-PCR法检测,hMLM1启动子区甲基化率分别为4.8%(2/42)和11.9%(5/42),启动子区甲基化与性别、年龄、病理类型和病理分级无明显的相关性。结论在脑胶质瘤中hMLM1基因启动子区甲基化少见,MSP法和限制性内切酶-PCR法是简单、灵敏的甲基化检测方法。

【Abstract】 Objective To investigate the status of methylation of hMLH1 promoter in glioma,and to compare the different methods for measurement of DNA mythelation.Methods Methylation of the hMLH1 promoter was measured in 42 glioma and 5 normal cerebral tissues by methylation specific PCR(MSP) and restriction endonucleases-PCR.Results The incidence of methylation in the hMLH1 promoter were 4.8%(2/42) by MSP and 11.9%(5/42) by restriction endonucleases-PCR.Methylation frequeney was not correlated with sex,age,pathological classification and stage.Conclusions Hypermethylation of hMlH1 promoter is rare in glioma. MSP and restriction endonucleases-PCR are easy and sensitive methods to measure DNA mythelation.
