TRAIL联合丝裂霉素对前列腺癌细胞DU145杀伤作用的实验研究
The effect of TRAIL combined with mitomycin C on apoptosis of human prostatic cancer cell DU145
【摘要】 目的研究肿瘤坏死因子相关的凋亡诱导配体(TRAIL)和丝裂霉素C(MMC)对前列腺癌细胞的杀伤作用及其协同作用。方法选用激素非依赖的前列腺癌细胞株DU145,将生长良好的细胞接种于96孔培养板,培养24 h后用MMC(25、50、100、200μg/mL)和TRAIL(10、50、125、250、500 ng/mL)分别/联合作用12 h,倒置纤维镜下观察细胞形态的变化,然后用MTT法测定细胞的生长抑制率。结果①细胞形态变化(50~500)ng/mL的TRAIL组、(50~200)μg/mL的MMC组、以及上述浓度的TRAIL和MMC联合用药组部分细胞变圆,从培养瓶壁脱落,核/浆比例增大,胞浆颗粒增多,胞核有皱缩变形,大小不均。上述改变的程度随着药物浓度的增加而加重。②细胞生长抑制率10 ng/mL TRAIL和25μg/mLMMC对DU145细胞的生长抑制率和不用药组比较无显著性差异(P>0.05);(50~500)ng/mL TRAIL和(50~200)μg/mLMMC对DU145的生长抑制率显著高于阴性对照组(P<0.05),而且随着TRAIL和MMC浓度的升高,生长抑制率显著增高(P<0.05);(50~500)ng/mL的TRAIL和(50~200)μg/mL的MMC分别联合作用对DU145细胞的抑制率均高于单独给药组(P<0.05);10 ng/mL的TRAIL单独用药对DU145细胞的抑制作用虽不显著(P>0.05),但和(50~200)μg/mL MMC联合用药均能加强后者对DU145细胞的抑制作用(P<0.05)。结论TRAIL和MMC在杀伤前列腺癌细胞时存在协同作用。
【Abstract】 Objective To investigate the effects of TNF-related apoptosis-inducing ligand(TRAIL) and mitomycin C(MMC) on the apoptosis of human prostatic cancer cell DU145. Methods Hormone-independent prostatic cancer cell line DU145 was selected in the study.100μl of DU145 cell suspension was added to each well of 96-well plate and cultured for 24 hours.Then the cells were treated with different doses of TRAIL,MMC,or TRAIL+MMC respectively for 12 hours.Death Rates of the cells were detected by MTT test. Results The death rates of groups treated with 10ng/ml TRAIL and 25μg/ml MMC were not significantly different from that of the negative control group(P>0.05).The death rates of groups treated with 50~500ng/ml TRAIL and 50~200μg/ml MMC decreased significantly when compared with the control groups(P<0.05).The Rates of groups treated with combination of 50~500ng/ml TRAIL and 50~200μg/ml MMC were significantly higher than those treated with the same doses of TRAIL and MMC lonely.Although the rates did not change significantly in the groups treated with 10ng/ml TRAIL(P>0.05),it could significantly increased the death rates of the groups treated with 50~200μg/ml MMC. Conclusion TRAIL can enhance the killing effect of MMC on prostatic cancer cell DU145.
