黄牛小腔卵泡卵母细胞体外受精及早期胚胎发育的影响因素

【摘要】 从屠宰场收集黄牛卵巢,取皮质深层卵母细胞进行体外成熟、体外受精和早期胚胎体外培养,分析影响其效果的因素。结果表明,在成熟培养液中添加促卵泡素(FSH)(10 IU/mL)、人绒毛膜促性腺激素(HCG)(20 IU/mL)和17β-雌二醇(17β-E2)(1μg/mL)对卵母细胞受精后早期胚胎发育能力有极显著的促进作用。等量牛卵泡液(BFF)与初生犊牛血清(NCS)效果差异不显著,以添加15%BFF为最宜。颗粒细胞与输卵管上皮细胞均能显著提高卵母细胞体外成熟受精后早期胚胎的发育率,颗粒细胞+输卵管上皮细胞对克服胚胎阻滞现象效果最显著。

【Abstract】 Factors influencing in vitro maturation,fertilization and embryo’s furtherdevelopment of bovine oocytes derived from interior ovary(IO) follicles was studied.The addition of follicle stimulating hormone(FSH)(10 IU/mL),HCG(20 IU/mL)(human chorionic gonadotropin) and 17β-E2(1 μg/mL)estradiol in maturation medium had a significant influence on the ability of fertilization of oocytes and embryo’s development of IO-oocytes(P<0.01).The similar effect wasobservedin the group of BFF(bovine follicle fluid)instead of NCS(neonate calf’s serum).The best doseof BFF added was 15 percent.The cultivation with monolayer granular cells(MGC) and monolayer oviduct epithelial cells(MOEC)sin maturation medium increased the developmental rate of 4-to 8-cellembryoss(P<(0.05))and morulae(P<0.01).Therefore,MGC+MOECis the best cultivated system.
