ISSR标记在侧耳属菌株分类学中的初步应用
The Prelimery Application of ISSR Marker in Taxonomy of Pleurotus Species
【摘要】 基于重复序列(TATG)4,设计了7条引物P0~P6,对侧耳属的22个菌株和1株双孢蘑菇菌株的基因组DNA进行重复序列间区序列(ISSR)PCR扩增。引物P0、P1在23个菌株上扩增共得到120条带。聚类分析结果显示,在0.71水平将以上侧耳菌株分为5组:阿魏蘑、白阿魏蘑、杂交阿魏蘑位于第2组;凤尾831、16号秀珍菇位于第3组;鲍鱼菇位于第4组;其他供试菌株位于第1组。利用引物P1对17号日本秀珍菇扩增得到了1条约1.3 kb片段,经克隆和测序分析,验证了重复序列(TATG)4的存在,并达到了微卫星基序的最低重复次数。
【Abstract】 Seven primers synthesized based on 16-mer oligonucleotide(TATG)4 were screened with the DNA from 22 Pleurotu strains and one Agaricus bisporus strain.120 bands were amplified by primer P0 and P1.Strains were classified into five groups by UPGMA.The 1.3 kb band of No.17 strain were obtained with 1.5% agarose gel and sequenced.The(TATG)4 motif was found and reached the teast limit of repetition of a motif.The Cluster-analysis result showed plentiful variability within closely related strains and suggested ISSR is useful in taxonomy of Pleurotus strains.
