CCR5亲和短肽抗肿瘤活性研究
Studying on Anti-tumor Activity of CC Chemokine Receptor 5-binding Peptide
【摘要】 目的探讨趋化因子受体CCR5亲和短肽(AFDWTFVPSLIL)对肿瘤生成作用的影响。方法通过MTT实验、细胞凋亡实验检测短肽对血管内皮细胞生长的影响,通过鸡胚绒毛尿囊膜(CAM)实验检测短肽对血管生成的影响,通过体内抑瘤实验检测短肽对实体瘤生长的作用。结果短肽能通过诱导人脐静脉血管内皮细胞的凋亡来抑制内皮细胞的生长;并能抑制CAM膜的血管生成。短肽也能在体内抑制小鼠B16黑色素瘤的生长活性,其抑瘤率达68.6%。结论初步证明短肽具有抗肿瘤生成作用,且这种作用可能是通过抑制肿瘤的血管生成来实现。
【Abstract】 Objective To study the effects of peptide(AFDWTFVPSLIL) on the growth of tumor.Method The action of peptide on vain endothelial cells (ECV304) was investigated through MTT assay,apoptosis assay;The activity of anti-tumor was tested through chick chorioallantoic membrane(CAM) angiogenesis assay and in vivo tumor suppression investigation.Results Peptide could inhibit the viability of human umbilical vein endothelial cells and the neovascularization of CAMs in a dose-dependent manner;Daily subcutaneous injection of peptide at a dosage of 10㎎/kg suppressed the growth of B16 murine melanoma by 68.6% as measured by average weight of tumor in mouse xenograft models of BALB/c mice.Conclusion Peptide had the anti-tumor activity which was acquired through inhibiting tumor angiogenesis.
