细胞外信号调节激酶1/2在压应力诱导血管平滑肌细胞增殖中的作用
Static Pressure Stimulating Vascular Smooth Muscle Cell Proliferation via the Extracellular Signal-regulated Protein Kinase 1/2 Pathway
【摘要】 目的探讨压应力对血管平滑肌细胞增殖的影响及其机制。方法用自行研制的压力培养箱加不同压力(大气压0 mmHg、正常血压120 mmHg、高血压180 mmHg和高血压危象血压240 mmHg)处理原代血管平滑肌细胞24 h。用MTT法和细胞计数法观察压应力对血管平滑肌细胞增殖的影响;用免疫印迹法检测细胞外信号调节激酶1/2的含量。结果与大气压组比较,正常血压压力组和高血压压力组的血管平滑肌细胞增殖速度和磷酸化细胞外信号调节激酶1/2的含量都明显增高(P<0.01),其中正常血压压力的作用又比高血压压力要明显(P<0.05);而高血压危象血压压力没有促增殖效应(P>0.05)。结论在一定范围内,压应力可通过细胞外信号调节激酶1/2途径促进血管平滑肌细胞增殖。
【Abstract】 Objective To investigate the effects of extracellular signal-regulated protein kinase 1/2(ERK1/2) on static pressure in vascular smooth muscle cell(VSMCs) proliferation.Methods VSMCs were respectively treated with atmospheric pressure 0 mmHg,normal blood pressure 120 mmHg,high blood pressure 180 mmHg,hypertension crisis blood pressure 240 mmHg in a self-manufactured pressure incubator for 24 hours.The proliferation of VSMCs was evaluated with cell count method and MTT method.Western Blot method was used to assay the protein amount of phosphatization-ERK1/2(p-ERK1/2).Results Static pressures stimulated VSMCs proliferation and ERK1/2 activation in normal blood pressure group and high blood pressure group, and those in the former group were more significant.However,too high pressure(hypertension crisis blood pressure 240 mmHg) had no effect on VSMC proliferation.Conclusion Static pressure stimulates smooth muscle cell proliferation via the ERK1/2 pathway.
