银杏叶提取物对树突状细胞和Th1/Th2细胞因子的影响
Effects of Ginkgo Extract on Function of Dendritic Cells and Th1/Th2 Cytokines
【摘要】 目的研究银杏叶提取物对不稳定型心绞痛患者(UAP)树突状细胞(DC)功能和Th1/Th2细胞因子的影响。方法将54例UAP分为常规治疗组(对照组,27例)和常规加银杏叶提取物组(银杏叶组,27例),分别于治疗前及治疗后4周取血,分离外周血单个核细胞,在含粒细胞、巨噬细胞、集落刺激因子和白介素4的培养条件下制备DC。用流式细胞仪检测DC表面CD86(B7-2)的表达;混合淋巴细胞反应(MLR)检测DC对同种异体T淋巴细胞的刺激能力;ELISA法测定血液中的细胞因子IFN-γ和IL-4;比浊法测定血C反应蛋白(CRP)。体外实验观察银杏内酯B对DC CD86表达的直接影响。结果与对照组比较,银杏叶提取物可进一步抑制DC功能(P<0.01),同时显著降低血CRP水平(P<0.05)和血IFN-γ浓度(P<0.01);银杏内酯B对DC CD86表达有直接的抑制作用(P<0.01)。结论银杏叶提取物对DC的抑制和由此对患者炎性反应的抑制可能是其治疗心绞痛的机制之一。
【Abstract】 Objective To investigate effects of Ginkgo extracton the function of dendritic cells(DC) and Th1/Th2 cytokines in patients with unstable angina pectoris(UAP).Methods 54 patientswith UAP were divided into two groups(control group 27 cases and Ginkgo group 27 cases) treated re-spectively with regular pharmacotherapy and regular pharmacotherapy plus Ginkgo extract.PBMC fromUAP patients(before and 4 weeks after the treatment) were incubated and induced to mature DC in thecompleted mediumcontaining GM-CSF and IL-4.The flowcytometric analysis were used to detect theexpression of co-stimulating factor CD86(B7-2)on DC.The stimulating capacity of DC was deter-mined in allogenic mixed lymphocyte reaction(MLR).ELISAwas used to analyze the blood levels of cy-tokines(IFN-γand IL-4),CRP level was also analyzed.The direct effect of Ginkgolide B on CD86expression on DC were also tested.Results When compared with pre-treatment,CD86 on DC wasmuch less expressed in both groups(P<0.01);the stimulating capacity of DC in MLR was lower;lower level of Th1 cytokine IFN-γand CRP were detected in both group after treatment for4 weeks(P<0.05,P<0.01).When compared with the control group,Ginkgo extractfurther inhibited the expres-sion of CD86 on DC and its stimulating capacity in MLR(P<0.01),and lowered the blood concentra-tion of IFN-γand CRP post treatment.In vitro,Ginkgolide B inhibited the expression of CD86 on DCdirectly.Conclusion Ginkgo extract may have an inhibitory effect on DC in patients with UAP.
