【摘要】 目的建立服用盐酸二甲双胍缓释片后人血清中盐酸二甲双胍浓度的测定方法。方法取血清样品0.5 m l,用35%高氯酸30μl沉淀蛋白,取上清液进样,色谱柱为L ichrospher C18柱(250 mm×4.6 mm,5μm),流动相为磷酸缓冲液-甲醇(60:40),流速1.0 m l.m in-1,检测波长232 nm。结果盐酸二甲双胍在0.05~2.51 mg.L-1范围内线性关系良好(r=0.9999),最低检测限为0.05 mg.L-1,日内、日间RSD均小于4%。结论该方法快速、灵敏、准确,可用于盐酸二甲双胍缓释片的人体药动学研究。更多还原

【Abstract】 OBJECTIVE To establish an HPLC method to determine the concentration of Metormin hydrochloride in human serum.METHODS 0.5 ml serum samples were precipitated with 35% perchloric acid and separated on Lichrospher C₁₈ column（250 mm×4.6 mm,5 μm）.The mobile phase was composed of buffer solution-methanol（60:40）with flow rate of 1.0 ml·min^-1 and detected at 232 nm.RESULTS The linear range was 0.050-2.51 mg·L^-1（r=0.9999）,and the limit of detection was 0.05 mg·L^-1.RSD of inter-day and intra-day assays were all less than 4%.CONCLUSION The method is stable and reliable for pharmacokinetic studies of Metormin hydrochloride.更多还原
