葛根素与胰蛋白酶相互作用研究
Study on the interaction between puerarin and trypsin
【摘要】 葛根素对胰蛋白酶有明显的抑制作用,D ixon作图法得出抑制常数Ki=8.15×10-5mol.L-1,抑制类型为非竞争性抑制。通过紫外吸收光谱,发现葛根素能改变胰蛋白酶的空间结构;由荧光光谱变化探索葛根素与胰蛋白酶的相互作用机理,发现荧光猝灭是能量转移引起的静态猝灭。
【Abstract】 The prohibitive function of puerarin over trypsin was studied.The value of Ki(8.15×10-5mol/L)was obtained by usingDixon graphical construction according to the ratio between 1/v and amount of inhibitor.It was found that inhibitor was a noncom-petitive.Utilizing ultraviolet spectroscopy,we found that puerarin can alter the molecular conformation of the trypsin.Utilizing fluo-rescence spectrophotometry we studied the mechanism of the interaction between puerarin and trypsin and found that fluorescencequenching was a static quenching due to nonradiative energy-transfer.
