犬骨髓间质干细胞体外定向诱导分化心肌细胞的实验研究
Experimental research on the myocardial cells selectively induced and differentiated in vitro from mesenchymal stem cells of canine bone marrow cells in vivo
【摘要】 目的建立骨髓间质干细胞体外诱导分化心肌细胞的方法。方法犬骨髓液10ml通过密度梯度离心法分离出间质干细胞,间质干细胞用5-氮胞苷诱导培养3~4周,进行心肌细胞特异性标志物测定及细胞超微结构的观察。结果犬骨髓间质干细胞经5-氮胞苷诱导培养3~4周后,免疫组化显示肌细胞特异性蛋白α-actin、Myosin、TropninⅠ阳性,心肌特异性肌钙蛋白I阳性。电镜观察细胞呈梭形,可见细胞肌丝与房性颗粒。结论密度梯度分离法分离出骨髓间质干细胞,用5-氮胞苷诱导后可分化为心肌细胞。此可为心肌梗死、心力衰竭与心律失常的移植及修复治疗提供理想的细胞材料。
【Abstract】 Objective To establish the method of inducing and differentiating myocardial cells from mesenchymal stem cells from bone marrow. Methods The bone marrow(10ml) was separated into mesenchymal stem cells through the density-gradient centrifugating method, and the solution was cuture- induced for 3 ̄4 weeks with 5-azacytidine. The cardiomyocyte specific indicator was measureed and the microstructure of the cell was observed. Results Immunohistochemistry showed α -actin, myosin and troponin Ⅰ of myocyte was positive, and troponinⅠof cardiomyocyte was positive. Conclusion Bone marroe mesenchymal stem cells can be separeated through the density-gradient centrifugating method. The cells can be differentiated into cardiomyocytes through cuture- inducement with 5-azacytidine. This research may provide the perfect cellular transplant and repair pproaches for the treatment of miocardial infarction, heart failure and arrhythmia.
