【摘要】 【目的】探讨基质金属蛋白酶9(MMP9)在大肠癌组织中的表达及其与大肠癌侵袭转移关系,基底膜(BM)的状态与大肠癌发生发展的关系。【方法】采用链霉卵白素过氧化物酶(SP法)免疫组织化学方法检测51例大肠癌1、5例肠腺瘤1、0例正常肠粘膜MMP9的表达与Ⅳ型胶原蛋白的表达情况。【结果】51例大肠癌MMP9阳性率84.3%,明显高于正常肠粘膜(20.0%)和肠腺瘤(26.7%)(P<0.05和P<0.01);MMP9的表达与大肠癌的分化程度、淋巴结转移及Dukes分期密切相关(P<0.05或P<0.01);正常肠粘膜和肠腺瘤组织中,基底膜Ⅳ型胶原连续完整,大肠癌基底膜Ⅳ型胶原分布间隔缺损者明显高于肠粘膜和肠腺瘤(P<0.01)。【结论】MMP9对肠癌基底膜破坏起着十分重要作用,且可作为预测大肠癌侵袭、转移的重要指标。更多还原

【Abstract】 【Objective】To explore the relationship between expression of matrix metalloproteinase 9(MMP9) and invasion,metastasis,and between the state of basement membrane(BM) and the progress of colonic carcinoma.【Methods】Fifty one cases of colorectal carcinomas,10 cases of normal mucosa,15 cases of adenomas were studied by using MMP9,type Ⅳ collagen immunohistochemical SP methods.【Results】Positive rate of MMP9 expression in the colorectal cancer specimens was 84.3%,which was significantly higher than that in the normal mucosa(20.0%,P<0.01),adenomas(26.7%,P<0.01).MMP9 positive expression related closely with differentiation,lymph node metastasis and Dukes staging(P<0.05 and P<0.01).Intact BM was seen in normal mucosa and adenomas.Fragmentary BM was more in adenocarcinomas than in normal mucosa,adenoma(P<0.01).BM disappearance was more easily seen in adenocarcinomas than in adenomas(P<0.01).【Conclusion】The results suggest that MMP9 may play an important role in the destruction of BM,and can be used to predict the invasion and metastasis of colorectal cancer.更多还原
