表皮生长因子受体与Dok1 PTB结构域结合关键位点的确定
The key binding site of EGFR and Dok1 PTB
【摘要】 目的确定表皮生长因子受体(EGFR)与Dok1PTB结构域结合的关键氨基酸残基位点。方法合成包含磷酸化的酪氨酸残基的EGFR胞内域的5个小肽片段,利用谷胱苷肽S转移酶共沉淀技术确定能够抑制Dok1PTB结构域与激活的EGFR结合的小肽。结果pY1086或pY1148小肽能抑制激活的EGFR与Dok1PTB结构域的结合。结论pY1086和pY1148是激活的EGFR与Dok1PTB结构域结合的关键位点。
【Abstract】 Aim: To study the EGFR key site binding to Dok1PTB domain.Methods: Five mini-peptides including phosphorated tyrosine origins of EGFR were used to identify the mini-peptide which could inhibit the binding of EGFR and Dok1PTB domain by GST pull down.Results: Mini-peptide pY1086 and pY1148 could inhibit the binding of activitied EGFR and Dok1PTB domain. Conclusion: EGFR tyrosine residues at 1086 and 1148 are essential for the binding between EGFR and Dok1PTB.
