藻酸双酯钠对急性脑缺血再灌注大鼠脑组织单胺类递质含量的影响

【摘要】 目的:观察藻酸双酯钠(PSS)对急性脑缺血再灌注大鼠大脑皮层、纹状体单胺类递质含量的影响。方法:大鼠60只,随机分为6组,每组10只。除假手术组,余5组(分别为阳性对照组、模型对照组和PSS大、中、小剂量组)均以线栓法制作急性大鼠中动脉脑缺血再灌注损伤模型。栓塞前30min,PSS大、中、小剂量组及阳性对照组分别腹腔注射PSS10mg/kg、5mg/kg和2.5mg/kg及尼莫地平3mg/kg,缺血1h再灌注4h后进行神经病学评分。另取大鼠60只,分组及处理方法同上,各组大鼠缺血2h再灌注1h后断头取脑,分离大脑皮层、纹状体,用荧光分光光度计测定5-羟色胺(5-HT)、多巴胺(DA)及去甲肾上腺素(NE)含量。结果:模型对照组大鼠出现神经运动功能障碍,PSS大、中、小剂量组及阳性对照组大鼠神经运动功能障碍均明显改善,PSS各剂量组间差异无统计学意义。与假手术组相比,模型对照组皮层及纹状体5-HT、DA、NE含量降低(P<0.01)。与模型对照组相比,PSS大、中、小剂量组及阳性对照组各单胺类递质含量均升高(P<0.05)。结论:PSS对缺血再灌注脑损伤的保护作用与其改善单胺类递质的代谢紊乱有关。

【Abstract】 Aim:To observe the effects of polysaccharide sulfate (PSS)on the contents of monoamine neurotransmitters in the cerebral cortex and striatum tissue of cerebral ischemia-reperfusion injury rats.Methods: Sixty rats were randomly and equally allocated into sham operation group, model group, high-,mediun- and low- dose PSS groups, and nimodipine group.Except for sham operation group, the other five groups were estabished ischemia-reperfusion model using the suture emboli method.The contents of 5-HT,NE,and DA in the cerebral cortex and striatum tissue of each group were determined by fluorospectrophotometry. Another 60 rats were grouped and treated as above, and the neural function were evaluated.Results:Compared with sham operation group, the neural function of model group were worse,and the contents of 5-HT,NE,and DA decreased significantly. Compared with model group, neural function were improved, and the contents of 5-HT,NE,and DA in PSS groups and nimodipine group increased significantly (P<0.05).Conclusion: The protective effects of PSS on the ischemia-reperfusion injury rats were related to the improvement of disturbance of monoamine neurotransmitter metabolism.
