脐血干细胞移植对帕金森病大鼠神经功能恢复的影响

【摘要】 目的:观察脐血干细胞移植对帕金森病(PD)大鼠神经功能恢复的影响。方法:PD大鼠模型分成实验组(n=10)和对照组(n=10)。将第三代脐血间充质干细胞(MSCs)用Hoechst33258标记后植入实验组大鼠纹状体内,对照组注射PBS。此后每周腹腔注射阿朴吗啡以观察神经功能恢复情况,并在2周,4周,8周用免疫荧光双标法检测MSCs的存活,迁移以及胶质纤维酸性蛋白(GFAP),神经元特异性烯醇化酶(NSE),酪氨酸羟化酶(TH)和突触素的表达。利用高效液相色谱电化学检测仪(HPLCECD)检测纹状体多巴胺含量。结果:脐血MSCs移植后大鼠的旋转行为与对照组相比有明显改善(P<0.05);MSCs可在大鼠脑内存活,随时间延长,迁移范围扩大,分布于纹状体,胼胝体和皮质;GFAP,NSE,TH都有表达,突触素无表达。多巴胺水平与对照组相比有明显提高(P<0.05)。结论:脐血MSCs有望成为治疗PD的种子细胞,为神经退行性变提供一种治疗方法。

【Abstract】 Aim: To explore the effect of human umbilical cord stem cell transplantation on neurological function recovery of PD rats.Methods:The PD rat models were allocated into the experimental group and the control group. The third generation MSCs were labeled by Hoechst33258,then were stereotactically transplanted into PD rat striatum, and the control group was given PBS. APO was injected to examine behavior of the rats in 2 groups each week. The brain tissue of PD rat was sectioned to identify the survival of MSCs and the expression of NSE,GFAP,TH, and synaptophysin under fluorescent microscope in the second, fouth, and eighth week. Dopamine in striatum was detected by high performance liquid chromatography. Results: The behavior of PD rat in experimetal group was improved obviously compared with control(P<0.05). Part of MSCs could survive, and move to callose,cortex with time and express NSE,GFAP, and TH. The contents of dopamine were far more increased than control(P<0.05).Conclusion: MSCs drived from human umbilical cord blood could provide a way to cure PD.
