转染人端粒酶RNA不同基因位点反义寡核苷酸的食管癌细胞在裸鼠体内生长情况观察
Observation on growth of esophageal cancer cells transfected by ASODN blocking different human telomerase RNA sites in nude mice
【摘要】 目的:观察不同基因位点的人端粒酶RNA(hTR)硫代修饰性反义寡核苷酸(ASODN)对裸鼠食管癌移植瘤的抑制作用。方法:40只裸小鼠随机等分为8组。将20μmol/L不同基因位点的hTRASODN(ASODN1、ASODN2、ASODN3、ASODN4及ASODN5)和无关序列寡核苷酸(N ODN)分别转染食管癌EC9706细胞,72h后采用裸鼠背部皮下注射的方法进行裸鼠体内成瘤实验,连续5周观察裸鼠体内食管癌移植瘤的生长情况。结果:空白对照和N ODN组最早长出肿瘤,ASODN4和ASODN5组次之,ASODN2、ASODN1和ASODN3组肿瘤长出最晚。35d时ASODN1组、ASODN2组、ASODN3组、ASODN4组、ASODN5组、N ODN组及空白对照组肿瘤体积(V/mm3)分别为450.1±58.2、581.1±69.7、237.7±140.5、1323.2±148.9、1187.3±258.0、1331.4±240.2及1476.2±131.8,前3组与后4组比较,差异有统计学意义(P<0.05)。与ASODN1、ASODN2及ASODN3组比较,ASODN4、ASODN5、N- ODN及空白对照组细胞异型性明显。结论:封闭hTR功能区的反义寡核苷酸可有效抑制裸鼠食管癌移植瘤的增殖及恶性表型的产生。
【Abstract】 Aim: To observe the inhibitory effects on esophageal transplanted tumors by ASODN blocking different hTR sites in nude mice. Methods: A total of 40 nude mice were randomly divided into 8 groups with 5 mice in every group.And mice of 5 groups were injected with esophageal carcinoma EC9706 cells transfected with 20 μmol/L ASODNs for 72 h (ASODN-1,ASODN-2,ASODN-3,ASODN-4, and ASODN-3 ) and N-ODN as experimental groups, with liposomes without ASODN as control group, and PBS as negative control group, respectively. The growth of nude mice transplanted tumors were observed for five weeks.Results: Transplanted tumors appeared firstly in control and N-ODN groups,secondly in ASODN-4 and ASODN-5 groups, and lastly appeared in ASODN-2,ASODN-1, and ASODN-3 groups.The tumor volumes (V/mm3) of ASODN-1, ASODN-2, ASODN-3, ASODN-4, ASODN-5, N-ODN, and control groups were 450.1±58.2, 581.1±69.7, 237.7±140.5, 1 323.2±148.9、1 187.3±258.0, 1 331.4±240.2, and 1 476.2±131.8, respectively. There was significant difference between the fomer three groups and the last four groups(P<0.05). Compared with ASODN-1,ASODN -2, and ASODN-3 groups, malignant phenotype of ASODN-4,ASODNGAA 5,N-ODN, and control groups were obvious.Conclusion: ASODN against the hTR function region could effectively inhibit proliferation and producing of malignant phenotype in nude mice transplanted tumors.
