尼莫地平对胶质瘤U251MG细胞生长的影响
Effect of nimodipine on growth of human glioblastoma U251MG cells
【摘要】 目的:研究钙通道阻滞剂尼莫地平(ND)对人脑多形性胶质母细胞瘤U251MG细胞生长的影响。方法:将U251MG细胞配成1×105ml-1的细胞悬液,接种孵育24h,细胞贴壁后吸除原培养基,分别加入含不同浓度rhT-GF-α(12.5pg/L、25.0pg/L、50.0pg/L)、不同浓度ND(1.0×10-6mol/L、1.0×10-5mol/L、1.0×10-4mol/L)加rhTGF-α(12.5pg/L)的无血清DMEM继续孵育3d。然后MTT法测定各组细胞的生长情况。以体积分数10%小牛血清为对照。结果:rhTGF-α对U251MG细胞有促生长增殖作用,且该作用呈一定的量-效依赖关系。不同浓度ND加12.5pg/L rhTGF-α联合作用后,细胞生长受抑,且抑制作用随ND浓度的增加而增强。结论:尼莫地平可能通过抑制细胞钙离子的内流与释放,阻断rhTGF-α对肿瘤细胞的生长刺激效应,从而抑制了肿瘤细胞生长增殖。
【Abstract】 Aim: To study the effect of the calcium channel antagonist nimodipine on the growth of U251MG,a human malignant glioma cell line.Methods:U251MG were cultured in various concentration of rhTGF-α(12.5 pg/L,25.0 pg/L,50.0 pg/L)only or in 12.5 pg/L rhTGF-α combined with ND(1.0×10~ -6 mol/L,1.0×10~ -5 mol/L or 1.0×10~ -4 mol/L) and serum-free medium, respectively.The growth of U251MG was observed using MTT analysis.Results: The TGF-α significantly enhanced the growth of U251MG cells in a concentration-effect dependent manner.Nimodipine mostly blocked the growth-stimulating effects of TGF-α on U251MG cells in a concentration-effect manner. Conclusion: Nimodipine can suppress the growth-stimulating effects of TGF-α,which suggests it could be used as a adjuvant therapeutic agent for human gliomas.
