【摘要】 应用鸡胚成纤维细胞(CEF)培养MDV-814株,经反复冻融、超声波处理、差速离心法纯化病毒,制成油乳剂灭活抗原;皮内多点注射法免疫健康家兔,获得高效价的免疫血清,经56℃、30min灭能,用硫酸铵沉降3次后,再用透析袋透洗,以除去部分NH4+和SO42-,最后用DEAE离子交换柱提取IgG。用IgG 2次标记金黄色葡萄球菌A蛋白(SPA)制成致敏的SPA菌悬液,用SPA协同凝集试验检测病鸡羽毛囊中MD特异性抗原。试验证明,上述方法的灵敏度是AGPT的48倍。整个试验只需几分钟即可完成,不需要复杂的仪器设备,具有操作简便,检测速度快,结果准确和便于现场应用等优点。更多还原

【Abstract】 Marek’s disease virus strain（MDV-814） was cultured in chicken embryo fiber（CEF） cells,treated with ultrasonic wave,purified at different speeds,in centrifugal pump,then MDVS was made as oil into Inactivated oil-emulsion antigen,which was used to immunize healthy rabbits to obtain antiserum to MDV.The antiserum was inactivated for 30 minutes at 56℃,the antibodies of the sera were purified by using ammonium sulfate precipitation followed by a dialysis process in dialysis bay to removing partial NH₄⁺ and SO₄^2-,and finally IgG was purified by DEAE-Cellulose Column.The SPA tagged twice by IgG,together with agglutination test,was used to detect the specific antigen of MDV in feather follicle.The method was rapid,accurate and needed no apparatus.it may be a useful technique for the diagnosis of avian Marek’s disease.更多还原
