【摘要】 为了建立烟草漂浮育苗旧苗盘残留物中烟草花叶病毒TMV的检测方法,以TMV病苗干根为材料,采用直接研磨法、液氮研磨法和浸泡研磨法制样,效果分别用三抗体夹心法(TAS-ELISA),TMV试纸条和枯斑寄主法检测灵敏度.结果表明,3种方法制备的样品都能用TAS-ELISA和试纸条检测出阳性,且3种制样方法间的差异小;3种制样方法制备的样品都具有侵染力,浸泡研磨法最强,直接研磨法次之,液氮研磨法最弱.TAS-ELISA,TMV试纸条和枯斑寄主接种的检测下限分别为干病根占健康根质量的0.01%,0.10%和0.10%.更多还原

【Abstract】 In order to develop detection methods of Tobacco mosaic virus in residue of tobacco roots on tray using in float seedling system,by using dry roots of TMV-infected tobacco float seedling,three sample prepare methods including direct grinding,liquid nitrogen grinding and dip grinding,and three detection methods sensitivity including TAS-ELISA,TMV-Imunostrip and local lesion assay are evaluated.Samples prepared by three methods can detect TMV positive and show no obvious difference between each other when detected by TAS-ELISA and TMV-Imunostrip,Samples prepared by three methods show TMV effective when inoculated into Nicotiana glutinosa,the local lesion number order is: dip grinding >direct grinding>liquid nitrogen grinding.The detection threshold of TAS-ELISA,TMV-Imunostrip and local lesion assay are 0.01%,0.10%,0.10% dry TMV-infected roots among healthy roots respectively.更多还原
