超声波辐照对大肠杆菌细胞膜的影响
Effect of Ultrasonic Radiation on Escherichia coli Membrane
【摘要】 为了解大肠杆菌经超声波作用后其细胞膜的变化状况,采用荧光染料二乙酸荧光素、罗丹明123、二氯荧光黄双乙酸盐对大肠杆菌样品染色,并通过荧光分光光度计、荧光显微镜和透射电镜检测、观察.结果显示:荧光探针测定法可用于测定大肠杆菌细胞膜的变化.在超声波频率为25 kHz、电功率为300W的条件下处理90 s后,细胞结构完整,膜双分子层变模糊,表面有破损成小孔的地方,内含物外渗.膜通透性增加12.7%,膜电位下降26.7%,胞内活性氧水平上升.
【Abstract】 Three fluorescent dyestuffs,namely FDA,Rh123 and DCFH-DA,were used to stain Escherichia coli(E.coli),and the fluorescence spectrophotometer,the fluorescence microscopy and the TEM were adopted to observe the membrane alteration of E.coli radiated by ultrasonic.The results indicate that fluorescent probe is feasible in the determination of the alteration of E.coli membrane,and that,after being radiated by ultrasonic at 300W and 25kHz for 90s,the cell maintains an integrated structure,but the double molecule layer of the membrane becomes blurry,with apertures on the surface and with the content material penetrating to the outside.Moreover, the membrane potential decreases by 26.7% while,in contrast,the membrane permeability increases by 12.7%,with the ROS level significantly improved.
