肺癌等不同疾病出现阴虚综合征时细胞因子基因表达谱的实验研究

【摘要】 目的研究肺癌、淋巴瘤、肺结核出现阴虚综合征时是否具有相同或相似的细胞因子基因表达变化规律,验证阴虚综合征的发病学机理是由于白细胞介素1(IL-1)、肿瘤坏死因子(TNF)等炎性细胞因子的基因表达增强、生物学活性相对升高引起细胞因子网络紊乱的理论研究结论。方法以外周血单个核细胞为材料,使用细胞因子基因芯片检测阴虚综合征细胞因子基因表达谱的改变。结果肺癌阴虚综合征时白细胞介素1β(IL-1β)、白细胞介素4(IL-4)、白细胞介素8(IL-8)和肿瘤坏死因子a(TNFa)的基因表达水平上调;淋巴瘤阴虚综合征时IL-8的基因表达水平上调;肺结核阴虚综合征时IL-1β、白细胞介素18(IL-18)和TNFa的基因表达水平上调。结论肺癌、淋巴瘤和肺结核伴有阴虚综合征时炎性细胞因子的基因表达水平上调,从细胞因子网络的角度,初步验证了阴虚综合征的发病学机理的理论研究结论。

【Abstract】 Objective To study whether to have the same or similar changing regularities of gene expression of cytokines among lung cancer,lymphoma and pulmonary tuberculosis with Yin-deficiency syndrome and to verify the preliminary conclusion that the pathogenesis of the Yin-deficiency syndrome is due to the disequilibrium status of cytokine networks resulted from the relative increase of the bioactivity of the pro_inflammatory cytokine like IL-1 and/or TNF and so on.Methods The changing characteristics of gene expression profile of cytokines were studied in peripheral blood mononeuclear cells samples with gene chip.Results The gene expression level of pro-inflammatory cytokines like IL-1β,IL-4,IL8,TNFa was up_regulated in lung cancer Yin_deficiency patient.The gene expression level of IL-8 was up_regulated in lymphoma Yin-deficiency patients;The gene expression level of IL-1β,IL-18,TNFa was up_regulated in pulmonary tuberculosis Yin_deficiency patients.Conclusions The experimental results showed that the gene expression level of multiple pro_inflammatory cytokines like IL-1β,IL-8,IL-18,TNFa and so on was up_regulated in Yin_deficiency patients.The results conformed to the theory research conclusion that the pathogenesis of Yin_deficiency syndrome is due to the disequilibrium status of cytokine networks resulted from the relative increase of the bioactivity of the pro_inflammatory cytokine like IL-1 and/or TNF and so on.
