低剂量双酚A与17β雌二醇对MCF-7细胞增殖作用的剂量效应关系及其联合作用探索
On single and combined estrogenic effects of bisphenol A and 17β-estradiol at low dose assessed using MCF-7 cell proliferation
【摘要】 采用MCF7细胞增殖实验研究了类雌激素双酚A(BPA)、17β雌二醇(E2)对MCF7细胞增殖效应的单独作用以及两者间联合作用的剂量效应关系.结果显示,BPA在10-10~10-3mol·L-1浓度范围内、E2在10-14~10-3mol·L-1浓度范围内的单独作用均存在低浓度促进、高浓度抑制的倒U型剂量效应关系曲线,体系中E2浓度分别为10-8、10-11、10-14mol·L-1情况下,BPA在10-3~10-9mol·L-1浓度范围内和E2的联合作用与其单独作用存在显著差异,呈现出独立作用模式.
【Abstract】 The single and combined estrogenic effects of exoestrogen bisphenol A (BPA) and natural hormone 17β-estradiol(E2) were studied by the proliferation of MCF-7 human breast cancer cell line. The results showed that the dose-effect relationships of bisphenol A and 17β-estradiol exhibited reversed U-shaped curves. Both the chemicals (BPA: 10 -10~10 -3 mol·L -1, E2:10 -10~10 -3 mol·L -1) stimulated cell proliferation at low doses and inhibited it at high doses. Compared with the dose-effect relationship of single BPA the presence of E2 led to a marked change in the effect of BPA (10 -9~10 -3 mol·L -1) , which exhibited an independent action mode of BPA and E2 on the proliferation of MCF-7 cells.
