混合植物甾醇琥珀酸酯化物高效液相色谱分析
Analysis and Determination of Phytosterol Hemisuccinate by HPLC
【摘要】 应用高效液相色谱法对混合植物甾醇琥珀酸酯化产物中豆甾醇琥珀酸单酯、β-谷甾醇琥珀酸单酯的含量进行了分析测定。色谱柱为HypersilODS反相柱(4.6mm×150mm,5μm),流动相为色谱纯甲醇,质量流量0.9mL/min,紫外检测波长为210nm,恒溶剂洗脱。实验结果表明,豆甾醇琥珀酸单酯、β-谷甾醇琥珀酸单酯、菜籽甾醇琥珀酸单酯和菜油甾醇琥珀酸单酯的分离效果较好,响应时间均在10min左右,面积归一法能够快速准确地用于该过程的分析测试。
【Abstract】 Analysis and determination of stigmasterol hemisuccinate and β-sitosterol hemisuccinate in phytosterol hemisuccinate were studied by HPLC. chromatographic separation was carried out using a Hypersil ODS column (4.6 mm×150 mm, 5 μm) under isocratic conditions with methanol as mobile phase at a flowrate of 0.9 mL/min. The operative wavelength was set at 210 nm.The experimental results showed that phytosterol hemisuccinate could be separated in 10 minutes and that the contents of stigmasterol hemisuccinate and β-sitosterol hemisuccinate could be determinated accurately by the peak area normalization method.
