六神丸逆转灯盏细辛抑制MCF-7细胞凋亡作用的实验研究
Experiment research of the reversed effect of Liushenwan on the inhibition effect of Erigeron breviscapus injection on tumor cell apoptosis
【摘要】 目的观察六神丸与灯盏细辛注射液对人乳腺癌MCF-7细胞的作用,并从细胞凋亡角度探讨其相互作用机制。方法将六神丸含药血清及灯盏细辛注射液作用于体外培养的人乳腺癌MCF-7细胞,检测其对细胞增殖的直接抑制作用,流式细胞仪检测其对细胞凋亡及对Caspase-3表达的作用。结果六神丸含药血清对MCF-7细胞的增殖均有直接抑制作用,24、48、72 h生长抑制率分别为41.23%、36.93%、36.82%;灯盏细辛注射液对MCF-7细胞的增殖均亦有直接抑制作用,24、48、72 h生长抑制率分别为27.09%、19.40%、25.58%;两者合用作用增强,24、487、2 h生长抑制率分别为67.89%、65.57%、68.31%。流式细胞仪检测证明,灯盏细辛注射液可抑制MCF-7细胞凋亡及Caspase-3表达,六神丸含药血清可促进MCF-7细胞凋亡及Caspase-3表达。两者合用,MCF-7细胞凋亡及Caspase-3表达较对照组增强,效果界于两者单用之间。结论六神丸可通过逆转灯盏细辛注射液抑制MCF-7细胞凋亡及Caspase-3的表达而表现出促MCF-7细胞凋亡的作用。
【Abstract】 Objective To investigate the effect of liu-shen-wan and Erigeron breviscapus injection to human MCF-7 cancer cells,and research the mutual act mechanism from cell apoptosis.Methods Liushenwan and Erigeron breviscapus injection were added to MCF-7 cells cultured.The direct inhibition effect of cell multiplication was detected.The effect of the express of Caspase-3 and cell apoptosis were detected by FCM.Results Both serum of Liushenwan and Erigeron breviscapus injection can directly inhibit the cell multiplication.The inhibition rate of 24th hour,48th hour and 72nd hour of Liushenwan are 41.23%,36.93%,36.82% respectively.The inhibition rate of 24th hour,48th hour and 72nd hour of Erigeron breviscapus injection are 27.09%,19.40%,25.58%.Combination of Liushenwan and Erigeron breviscapus injection can get better effect than use one alone.The inhibition rate of 24th hour,48th hour and 72nd hour are 67.89%,65.57%,68.31% respectively.Proved by FCM,Erigeron breviscapus injection can restrict the express of Caspase-3 and cell apoptosis,but the serum of Liushenwan can promote the express of Caspase-3 and cell apoptosis.The effect of combination of Liushenwan and Erigeron breviscapus injection was superior to that in control group.Conclusion Liushenwan can reverse the Erigeron breviscapus injection’s restraint effect of the express of Caspase-3 and cell apoptosis,and it can promote the cell apoptosis of MCF-7.
