螺内酯和氯沙坦及二者合用对大鼠急性心肌梗死早期愈合的影响
Effects of combination of spironolactone and losartan on myocardial healing during the early stage of acute myocardial infarction in rats
【摘要】 目的探讨螺内酯和氯沙坦对急性心肌梗死(AMI)大鼠早期愈合的影响。方法随机将60只大鼠分为假手术组(Sham组n=12)与AMI组(n=48)。通过结扎左冠状动脉前降支造成AMI大鼠模型。再将术后72h存活大鼠随机分为AMI组、AMI+螺内酯组(S组)、AMI+氯沙坦组(L组)、AMI+螺内酯+氯沙坦组(SL组),用氯胺T法观察梗死区与梗死周围非梗死区的胶原含量,观察各组梗死周围非梗死区毛细血管密度,利用逆转录-PCR技术的方法,观察AMI早期大鼠心肌基质金属蛋白酶-13(MMP-13)mRNA的表达。结果组间梗死区与梗死周围区胶原含量差异并无显著性;AMI用药组梗死周围区毛细血管密度均高于AMI组;螺内酯和氯沙坦及2者合用组在心肌梗死后2周时MMP-13mRNA的表达均明显升高。结论螺内酯和氯沙坦及2者合用并不延缓心肌梗死后早期组织修复。
【Abstract】 Objective To evaluate the effects of combination of spironolactone and losartan on myocardial healing during the early stage of acute myocardial infarction in rats.Methods Sixty rats were randomly divided into sham group (n=12) and AMI group(n=48).LV in farction in rats was produced by ligation of left anterior descending coronary artery.The sham group did not undergo ligation of left anterior descending coronary artery but were threaded as well as the others. After 72 hours, stable AMI rats were randomly divided into AMI group, AMI +Spironolactone group, AMI +losartan group,AMI+Spironolactone + losartan group.The collogen contents were measurred by hydroxyproline levels in both infarcted and non-infarcted zone. And MMP 13 mRNA were detected by RT-PCR in non infarct zone. A comparison of capillary density has been done in every group.Results There was no significant difference for collogen contents in both non-infracted zone and infracted zone during early stage after myocardial infarction in rats among every AMI groups and AMI treated groups;the capillary density in non-infracted zone after AMI rats with treated in ① and ② week were significantly higher; The express of MMP 13 mRNA with combination of spironolactone and losartan in non-infracted zone two weeks were singnificantly higher than that in others groups.Conclusion Combination of spironolactone and losartan did not delay healing during early stage after AMI in rats.
