P21WAF1/CIP1在大鼠肾上腺细胞培养中的表达及其意义
Expression and Significance of P21 in Rat’s Adrenocortical Cells in Vitro
【摘要】 目的:探讨细胞周期依赖性激酶抑制剂(CDI)P21在大鼠肾上腺细胞培养过程中的表达及其意义。方法:应用SP免疫组化法,检测细胞周期依赖性激酶抑制剂P21在大鼠肾上腺细胞培养不同时期表达的强度变化,同时测定肾上腺细胞分泌皮质酮功能的动态变化。结果:在肾上腺细胞体外培养过程中,第3,5,7天,P21蛋白表达逐渐减弱,第9,11天与第7天相比,P21蛋白表达重新增强(P<0.05),皮质酮浓度测定显示在第7天皮质酮浓度达到最高峰,随后逐渐下降,与P21蛋白表达的变化趋势一致。结论:肾上腺细胞体外培养在第7天肾上腺细胞增殖最旺盛,而且肾上腺分泌皮质酮达到最高峰。
【Abstract】 Objective: To explore the expression and significance of cyclin dependent kinase inhibitor P21 in adrenocortical cells of rats in vitro.Methods: Immunohistochemical methods were adopted,expressing strengths of P21 were examined in cell culture in different phase,and meanwhile,corticosterone concentrations were dynamically measured.Results: Expression of P21 in adrenocortical cells was decreasing at the third,fifth or seventh day,in contrast to the ninth or eleventh day when P21 gradually increased(P<0.05).Corticosterone measurements showed corticosterone concentration was at peak in the seventh day,and gradually decreased.Changes of P21 expression degree were consistent with those of corticosterone concentrations.Conclusion: Adrenocortical cells were the most proliferative in vitro and excreting corticosterone was at the peak in the seventh day.
