云芝产漆酶发酵条件的研究
Research on the Conditions for Laccase Production from Trametes versicolor
【摘要】 研究了培养基初始pH值、通气量、温度、表面活性剂、诱导剂、金属离子对云芝(Trametes versi-color)HS-03产漆酶发酵条件的影响.研究发现培养基的初始pH、培养温度、表面活性剂、诱导剂和金属离子等因子对该菌漆酶产量均有明显的影响.结果表明,云芝摇瓶产漆酶的最佳培养条件为培养基初始pH为3.5,转速150 r/min,培养温度28℃,另加1 mL/L吐温-80,1 mL/L愈创木酚,终浓度为0.5 mmol/L的Cu2+.在该条件下,该菌的漆酶酶活力达到291 U/L,比优化前提高了8倍,同时产酶高锋也提前了5 d.
【Abstract】 Effects of initial pH,aeration rate,temperature,surface-active detergent,inducer and metal ions on laccase production from Trametes versicolor HS-03 were studied.The results indicated that the optimal fermentation conditions were as follows:initial pH 3.5 of the medium,rotative velocity 150?r/min,at 28?℃,Tween-80 1.0?mL/L,guaiacol 1.0?g/L and CuSO4·5H2O 0.5?mmol/L.On this condition,what the laccase activity reached 291?U/L is higher 8 times than in the before of optimizing and enzyme activity was ahead of 5?day for peak.
