重组鲑鱼降钙素融合蛋白工程菌发酵工艺研究
Study on fermentation process of fusion protein of recombinant salmon calcitonin expressing E.coli
【摘要】 为研究大肠杆菌表达重组鲑鱼降钙素融合蛋白(GST-sCT-Gly)工程菌的发酵工艺,采用瑞士比欧生物工程公司40 L发酵罐发酵,对影响工程菌生长和目的蛋白的表达条件进行优化;在优化条件下,以40 L发酵罐发酵工程菌,菌体收获量可达到57 g/L,目的蛋白表达量占总蛋白的23.5%;结果表明,研究确定的发酵工艺可以提高工程菌收获量及目的蛋白的表达量,且稳定可靠,可应用于生产。
【Abstract】 Objective:To develop the best fermentation process on E.coli expressing fusion protein of recombinant salmon calcitonin GST-sCT-Gly).Methods:The expression pf recombinant fusion protein was analyzed to optimize the composition of medium,induction time and other factors in the process of fermentation.Results:Under the estabilished condition,57 g of wet bacteria per liter was obtained and the derivative of recombinant fusion protein was about 23.5% of total protein on host.Conclusion:The established fermentative technique increases the collection efficiency and expression of recombinant fusion protein.The fermentation process was steady,and able to be used in pilot production.
