【摘要】 选用引物JYF5/JYR5对湖北疑似柑橘溃疡病标本进行了PCR鉴定和确证,同时改进了DNA抽提方法,优化了PCR条件,并与南京柑橘溃疡病菌株进行分子克隆比较。结果表明,南京菌株NJ-XC分离物扩增片段与Silva等报道的相应序列相似性为99%(411/413);湖北标本菌株HB-X0扩增片段与报道的相应序列相似性为100%(413/413),二者存在一定的差异。此外发现引物JYF5/JYR5能扩增叶表的根癌土壤杆菌,其片段大小为575bp。更多还原

【Abstract】 In this paper,a citrus sample from Hubei was identified by PCR Assay.At the same time,the method of DNA extraction was developed,and it was cloned and sequenced to make a comparison with Xac from Nanjing.The result showed that the two different strains had certain discrepancy in sequences.The comparability between the Nanjing sequence and that reported by Silva(2002)was 99 percent(411/413);the comparability between the Hubei sequence and the reported was 100 percent(413/413).Under the same primer,the amplified fragments of and unknown stain derived from the disease spots isolation showed high comparability of 93 percent(484/515)with one section of Agrobacterium tumefaciens sequences(575bp).更多还原
