马铃薯多酚氧化酶的提取纯化条件对其活性影响的研究

【摘要】 采用丙酮法、缓冲溶液法对马铃薯中多酚氧化酶(PPO)进行了提取后,采用硫酸铵分级沉淀法、凝胶层析法及分级沉淀和层析相结合的方法对多酚氧化酶进行了纯化,然后在不同的工艺条件下对PPO活性进行了研究。实验结果表明硫酸铵沉淀和凝胶层析相结合的方法能够大大提高PPO的活性,且在最佳pH值7.0和最适温度30℃的条件下PPO的活性达到1560 U,并用高效毛细管电泳法对所提取的酶进行了分析。

【Abstract】 After polyphenol oxidase(PPO) was extracted from potato with the acetone powder method,or buffer solution method,it was purified in the ways of ammonium sulfate precipitation,column chromatography and the combination of two methods,and the effects of different conditions on the activity of PPO were studied in this paper.The results showed that the combination method of ammonium sulfate precipitation and column chromatography could greatly improve the activity of PPO,with optimum pH value 7.0 and temperature 30℃.The activity could be 1560 U under the optimum condition,HPCE was also used to analyze the purified PPO.
