【摘要】 目的用高效液相色谱-蒸发光散色检测器(HPLC-ELSD)对一组氨基糖苷抗生素的响应因子的一致性进行测定,进一步探讨建立一些氨基糖苷抗生素,包括多组分氨基糖苷抗生素中各组分相对含量的测定方法。方法用HPLC-ELSD法测定K anam ycin A、B,抗生素G-418,抗生素JI-20B,抗生素TBJ-B和小诺霉素。HPLC条件:柱ODS C18;柱温40℃;流动相0.05m o.lL-1三氟乙酸:甲醇(93∶7);流速0.5mL.m in-1。ELSD条件:氮气体流速1.3L.m in-1;雾化温度70℃;蒸发器温度110℃。结果在选定的色谱条件下,测出一组氨基糖苷抗生素的线性方程为:K anam ycin A:Y=1.312X+5.567 r=0.9999;K anam ycin B:Y=1.329X+5.532 r=0.9991;抗生素G-418:Y=1.334X+5.3 r=0.9995;抗生素JI-20B:Y=1.3X+5.367 r=0.9994;抗生素TP J-B:Y=1.283X+5.36 r=0.9991;小诺霉素:Y=1.317X+5.345 r=0.9997。经t检验,各线性方程间无显著差异。结论所测定的一组氨基糖苷抗生素在HPLC-ELSD中的响应因子一致。更多还原

【Abstract】 OBJECTIVE An investigation on the response factors of various aminoglucoside antibiotics in HPLC-ELSD（evaporative light-scatter detector）was performed to probe the possibility to establish a method to determine the contents of each related materials simultaneously.METHODS Kanamycin A,Kanamycin B,G418,JI-20B,TPJ-B and Sagamicin were detdcted with HPLC-ELSD.The ODS C₁₈ Column was apolied at the temperature of 40℃.The mobile phase was 0.05mol·L^-1 trifluoroacetic acid-methanol（93∶7）and flowed at 0.5mL·min^-1.For the detector,the flow rate of carrier gas was 1.3L·min^-1;The nebulization temperature was set at 70℃;The evaporator temperature was 110℃.RESULTS The linear regression equation:Kanamycin A:Y=1.312X+5.567 r=0.9999;Kanamycin B:Y=1.329X+5.532 r=0.9991;G-418:Y=1.334X+5.3 r=0.9995;JI-20B:Y=1.3X+5.367 r=0.9994;TPJ-B:Y=1.283X+5.36 r=0.9991;Saganicin:Y=1.317X+5.345 r=0.9997.CONCLUSION Various aminoglucoside antibiotics detected in HPLC-ELSD can give the same responsefactors.And it is possible to develop a method to determine the contents of related aminoglucosides simultaneously更多还原
