小鼠成骨转录因子Osterix的克隆及其功能的初步研究
Cloning and study of mouse osteogensis-related transcription factor Osterix
【摘要】 Osterix是成骨细胞分化终末期一种重要的转录因子,决定多种骨细胞标志蛋白的表达,但其作用的具体分子机制至今尚不清楚。用BMP-2诱导小鼠成肌细胞C2C12向成骨细胞分化,RT-PCR的方法是从分化了的C2C12中克隆到小鼠Osterix基因,并插入动物细胞表达载体pCDNA3.1中,用脂质体法转染C2C12细胞,发现能够促进该细胞的骨化。这证明已经成功克隆到Osterix基因,为进一步研究其分子作用机制打下基础。
【Abstract】 Osterix is an important transcription factor during the process of osteogenesis,it determine the exist of many differenciation markers of osteoblasts,yet the molecular mechanism is unknown.C2C12 cells was induced by adding BMP-2 into the culture medium,RT-PCR was used to amplified the Osterix gene,then it was inset into the multicloning site of eukaryotic expression plasmid pcDNA 3.1,the recombinant vector was tranducted into C2C12 cells.It was found that overexpression of Osterix may result in the expression ALP in C2C12 cells.
