天麻提取物对小鼠巨噬细胞吞噬功能和肝肾功能的影响
Effects of Gastrodia elata BI(GE).Extract on macrophages,hepatorenal function in mice
【摘要】 目的观察天麻提取物对小鼠巨噬细胞吞噬功能及肝肾功能的影响。方法70%乙醇加热回流法制备天麻提取物,高效液相色谱法测定天麻素含量,并用紫外分光光度法测量,绘制天麻素含量-吸光度曲线;小鼠分为实验组与对照组,分别以天麻提取物和生理盐水灌胃15天,检测小鼠腹腔巨噬细胞吞噬功能及血清谷丙转氨酶(ALT)和肌酐(Cr)水平。结果本实验所获天麻提取物中天麻素含量为1.6mg/ml,紫外分光光度法测定结果显示,天麻素含量在100~300μg/ml范围内与吸光度(A224nm)呈线性关系(天麻素含量Cμg/ml=-668.317+286.000×A224nm,t=5.690,P<0.05);天麻实验组吞噬率与吞噬指数均显著增高(P均<0.001);两组血清ALT和Cr含量差异无显著性(P均>0.05)。结论采用70%乙醇加热回流法制备天麻提取物,用紫外分光光度法测定其中天麻素含量,方法简单易行,成本低廉,且该提取物对小鼠肝肾功能无毒副作用,同时能增强小鼠巨噬细胞吞噬功能。
【Abstract】 Objective To explore the influence of Gastrodia elata BI.(GE)extract on the functions of macrophages,liver and kidney.Methods The extract was prepared by 70% ethyl alcohol back-flowing method.High-performance liquid chromatography(HPLC)and ultraviolet absorption spectrophotometry were used to examine the content of gastrodin.The mice were given the extract for 15 days,the phagocytic functions of macrophages,blood glutamicpyruvic transaminase(ALT)and creatinine(Cr)were detected.Results The extract contented 1.6mg/ml gastrodin.Ultraviolet absorption curve was μg/ml=-688.317+286.000×A 224nm(P<0.05).The phagocytic functions of macrophages was increased markedly in experimental mice.Blood ALT and Cr were not different between experimental group and control group(P>0.05).Conclusion 70% ethyl alcohol back-flowing method for extracting effective components of GE and ultraviolet absorption spectrophotometry for detecting gastrodin content are simple,low-priced and practical.The extract can activate mice’s macrophages,and has not hepatorenal toxicity.
