槲皮素对药物致心肌细胞损伤的保护作用

【摘要】 目的:探讨槲皮素(QUE)对柔红霉素(DNR)诱导的体外培养原代SD乳鼠心肌细胞损伤的保护作用。方法:分离培养心肌细胞,分为空白对照组、DNR组、DNR+QUE组、QUE组,孵育24h,显微镜下观察心肌细胞形态,检测培养介质中乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)、脂质过氧化产物丙二醛(MDA)的含量。结果:与DNR组比,DNR+QUE组脱落细胞减少,细胞碎片减少,LDH和MDA含量降低(P<0.05),SOD活性增高(P<0.05),且有剂量效应关系。结论:25~100μmol/L浓度范围的槲皮素能减轻柔红霉素对心肌细胞的损伤,具有细胞保护作用,机制可能与其抗氧化作用有关。

【Abstract】 Objective:To investigate the protective effects of quercetin on cultured SD neonate rat cardiomyocyte injury induced by daunorubicin. Methods:Cultured cardiomyocytes were divided into blank group, DNR group,DNR+QUE group and QUE group.After preincubation of cardiomyocytes for 24 hours,cytoprotection of quercetin was assessed by cell morphology,extracellular LDH,SOD and MDA.Results:Compared with DNR group,the shedded cardiomyocytes and the cell debris of DNR+QUE group was decreased,the content of LDH and MDA lowered(P<0.05)and the activity of SOD increased(P<0.05)with a dose-effect correlation.Conclusions:Quercetin with concentration between 25~100μmol/L can lessen cardiomyocyte injury induced by daunorubicin.Such cytoprotection at may be due to its antioxidation.
