蒽酮一硫酸法测定酵母中海藻糖的含量

【摘要】 本文对酵母中海藻糖的含量测定进行了研究,确定了用0.2%蒽酮-硫酸溶液(料液比1:4)测定海藻糖的条件:吸收波长为590nm;加热时间为2min;样品显色后应在30min内测定。在冰水浴15mL0.5mol/L三氯乙酸溶液条件下从活性干酵母中提取海藻糖的次数为八次,每次30min。八次提取共从1g燕山低糖即发干酵母(水份:3.63%)中获得了149.42mg海藻糖。

【Abstract】 The determine content of trehalose in yeast was studied in the article. The conditions of determination of trehalose content by 0.2% Anthrone-Sulphuric acid colorimetric method (the proportion of extraction is l:4) is as follows: absorption peak is 590nm,time of heating is 2min,the numerical value was determined in 30min after coloration. The times of extraction trehalose from dried active baker’s yeast are 8 times and every time 30min under 0℃, 15mL 0.5mol/L Cupric acetate. Trehalose(149.42mg) was obtained from 1g Yanshan instant dry yeast (the moisture is 3.63%) after extraction 8 times.
