【摘要】 目的探讨重组人生长激素(r-hGH)对人乳腺癌细胞株MCF-7(MCF-7)有无促进增殖和分化作用,以及r-hGH与5-氟尿嘧啶(5-FU)联合应用对MCF-7的化疗协调作用。方法以不同浓度r-hGH,以及不同浓度的r-hGH联合不同浓度的5-FU分别加入处于对数生长期的MCF-7中进行体外培养,48h后流式细胞仪检测细胞周期的变化,与对照组进行比较。结果①不同浓度的r-hGH与MCF-7作用后,其细胞周期的G0~G1期、S期、G2~M期的细胞百分比变化,差异均无统计学意义(P>0.05);②不同浓度的5-FU与MCF-7作用后,其细胞周期的G0~G1期细胞百分比显著增高(P<0.05),S期显著下降(P<0·01),G2~M期差异无统计学意义(P>0.05);③不同浓度的r-hGH联合不同浓度的5-FU与MCF-7作用后,其细胞周期的G0~G1期细胞百分比显著增高(P<0.01),S期和G2~M显著降低(P<0.01,P<0.05)。结论单独应用r-hGH对MCF-7的细胞周期无明显影响;r-hGH与5-FU联合应用对MCF-7的化疗具有协调作用,r-hGH能增强5-FU对MCF-7的化疗效果。更多还原

【Abstract】 Objective To study effect of recombinant human growth hormone (r-hGH) on cell cycle of a human breast cancer cell line MCF-7 , and study the effect of r-hGH plus 5-FU on the effect of chemotherapy in MCF-7.Methods MCF-7 cells were cultured for 48h in RPMI 1640 with r-hGH for different concentrations and with 5-FU and r-hGH for different concentrations , then cells were gathered and evaluated for cell cycle by flow cytometry (FCM).Results ①Compared with the control group , r-hGH had no significant effect on the percentage of G0～G1 , S and G2～M phase of MCF-7 (P>0.05).②Addition of 5-FU to the culture raised the percentage of G0～G1 phase significantly (P<0.01) and reduced S phase significantly (P<0.05), and did not change the percentage of G2～M phase significantly (P>0.05).③Compared with the control group ,5-FU plus r-hGH raised the percentage of G0～G1 phase significantly (P<0.01) and reduced S , G2～M phase significantly (P<0.05).Conclusion The r-hGH alone might not have effect on the cell cycle of MCF-7 cells . In vitro r-hGH might promote the sensitivity of chemotherapy of 5-FU to MCF-7 cells.更多还原
