健胃愈疡颗粒对胃溃疡大鼠胃组织核因子-κB的活化和IL-8表达的影响

【摘要】 目的:探讨大鼠胃溃疡胃组织核因子-κB(NF-κB)的活化及白细胞介素-8(IL-8)表达在健胃愈疡颗粒(JWYY)胃黏膜保护机制中的作用。方法:将56只SD大鼠随机分为正常组、假手术组、模型组、JWYY组,改良Okabe法建立大鼠乙酸性胃溃疡模型,造模后第8天和第16天分别应用原位杂交法、免疫组化学法检测NF-κBmRNA及其蛋白的表达和IL-8蛋白的表达。结果:造模第8天,模型组NF-κBmRNA及其蛋白明显从细胞质移位于细胞核,与正常组(0.41±0.17、0.29±0.11)、假手术组(0.54±0.15、0.38±0.15)比较其核阳性反应的细胞密度增加(5.92±0.61、9.13±1.21,均P<0.01),IL-8表达增加(58.95±7.34 vs 9.55±1.58、11.00±2.21,均P<0.01);应用JWYY,NF-κBmRNA及其蛋白的核移位被明显抑制,阳性反应细胞密度显著减低(3.77±0.59、5.37±0.99,均P<0.01),IL-8表达亦显著减低(22.34±8.39,P<0.01)。模型组第16天与第8天相比NF-κBmRNA及其蛋白部分从胞核移出细胞质,其核阳性反应的细胞密度显著减低(2.42±0.51、4.18±0.94,均P<0.01),IL-8表达亦减低(15.72±2.67,P<0.01);JWYY组NF-κBmRNA及其蛋白和IL-8表达(1.69±0.11、2.96±0.46、11.35±2.54)均不及模型组(分别P<0.05、P<0.05、P<0.01)。NF-κBmRNA及其蛋白表达均与IL-8的表达呈正相关(r=0.809、r=0.808,均P<0.01)。结论:JWYY抑制大鼠胃溃疡NF-κB的活化进而抑制炎症反应可能是其胃黏膜保护作用的机制之一。

【Abstract】 Objective:To investigate the effect of Jianweiyuyang granules(JWYY) on activation of nuclear-κB(NF-κB) and the expression of IL-8 in rats with gastric ulcer,and to discuss the probable mechanism of its gastric mucosa protective effect.Methods:56 SD rats were randomly divided into 4 groups: normal group,sham operation group,model group,and JWYY group.The gastric ulcer was induced in the rats by acetic acid according Okabe’s method with some modification.Immunohistochemistry(IH) and In Situ Hybridization Histochemistry(ISH) were used to detect the expression of NF-κBmRNA and its protein as well as IL-8 protein in the stomach.Result:The 8th day after the model was made,NF-κBmRNA and its protein were significantly translocated from the cytoplasm into the nucleus,however,NF-κBmRNA and its protein remained in the cytoplasm of gastric epithelium in normal and sham operation group.Compared to the normal group(0.41±0.17,0.29±0.11)、sham operation group(0.54±0.15,0.38±0.15),their cell density of positive reaction in nucleus increased(5.92±0.61,9.13±1.21,all P<0.01),so did IL-8 protein(58.95±7.34 vs 9.55±1.58,11.00±2.21,both P<0.01).NF-κBmRNA and its protein significantly shifted out from nucleus to cytoplasm on the 16th days.Cell density of positive reaction was observed decreasing in NF-κBmRNA and its protein(2.42±0.51,4.18±0.94,both P<0.01)as well as IL-8(15.72±2.67,P<0.01).The expression of NF-κBmRNA and its protein as well as IL-8 in JWYY group(1.69±0.11,2.96±0.46,11.35±2.54) were significantly lower than those of the model group( P<0.05,P<0.05,P<0.01).There was a significant positive correlation between expression of NF-κBmRNA,its protein and IL-8(r=0.809,r= 0.808,both P<0.01).Conclusion:The activation of NF-κB and the following inflammatory reaction may be one of the pathogenesis of acetic acid induced-gastric ulcer.JWYY inhibits the inflammatory reaction by inhibiting the NF-κB activation,which may be one of the mechanisms of its gastric mucosa-protective effect.
