L-亮氨酸发酵种子培养试验
Study on seed culture for L-leucine fermentation
【摘要】 应用正交设计试验法确定L-亮氨酸发酵的摇瓶种子培养基组成,应用单因素试验法研究种子培养条件。试验结果显示,以培养基初始pH 7.0、500 mL圆底三角瓶装液量50 mL、摇瓶转速为220 r/m in,28℃恒温振荡培养36h的种子接种量为10%,摇瓶分批发酵72 h产L-亮氨酸16.52 g/L.
【Abstract】 The seed medium ingredient in shake flask of L-leucine fermentation was confirmed by mean of orthogonal design experiment.The seed culture conditions was tested through single factor experiment.The results showed that the production of L-leucine reached 16.52g/L after flask-shaking batch fermentation for 72 hours when the seed culture conditions as follow,initial pH value in medium was 7.0,50mL medium in 500mL flask,shaking speed 220r/min,shaking culture temperature 28℃ for 36 hours and seed inoculation rate 10%.
