MHCⅠ类分子-抗原肽单链三聚体的制备及其应用
Preparation of MHC class Ⅰ-antigenic peptide single-chain trimer and its application in immunotherapy for cancers
【摘要】 主要组织相容性复合物(MHC)Ⅰ类分子-抗原肽单链三聚体(SCT)由MHCⅠ类分子的重链、β2微球蛋白及抗原肽分子用接头相连接折叠形成。SCT能有效地维持其共价结构,并稳定地在细胞表面表达,能更有效地激活抗原特异性的CD8+细胞毒性T淋巴细胞,从而能更有效地清除病原体感染和恶性肿瘤细胞。
【Abstract】 Major histocompatibility complex(MHC)classⅠmolecules-antigenic peptide single chain trimer(SCT)consists of an antingenic peptide-spacer-β2-microglobulin(β2m)-spacer-H chain.Recently,it is demonstrated that SCTs can efficiently maintain their covalent structure,and they are unusually stable at the cell surface.SCT can efficiently induce the generation of antigenic peptide-specific CD8+ cytotoxic T lymphocyte response,therefore they can play important roles in the eradication of infec- tion,particularly in viral infection and tumor cells.
