BMP-2促进鼠胚成骨细胞VEGF mRNA表达的时间依赖性效应
Time-dependent Effect of rhBMP-2 Promoting the VEGF Expression in Fetal Mouse Osteoblasts
【摘要】 目的探讨BMP-2促进鼠胚成骨细胞VEGF表达的时间规律。方法取19d胎龄的小鼠胚胎颅骨成骨细胞进行体外培养,采用RT-PCR法检测相同剂量BMP-2(300ng/ml)不同作用时间下对成骨细胞VEGFmRNA表达的影响。结果RT-PCR法检测表明,BMP-2不同作用时间点VEGFmRNA的表达量不同,呈双时相。6h时明显升高,约为对照组的2·6倍(P<0·05)。VEGFmRNA的第2个峰值出现在36h左右,约为对照组的2倍。48h时,VEGFmRNA的相对水平仍然保持较高水平。结论rhBMP-2对VEGFmRNA表达的促进作用呈时间依赖关系,分别在6h和36h出现峰值。
【Abstract】 Objective To investigate the time pattern of vascular endothelial growth factor (VEGF) expression promoted by recombinant human bone morphogenetic protein-2 (rhBMP-2) in fetal mouse osteoblasts.Methods Calvaria osteoblasts of 19-day fetal mouse were cultured and the effect of rhBMP-2 on VEGF mRNA and protein expression at different times in fetal mouse osteoblasts were observed with reverse transcription-polymerase chain reaction (RT-PCR).Results The levels of VEGF mRNA were increased in an apparent biphasic manner with a maximum stimulation (about 2 times over the control,P<0.05) of both mRNA species observed at 300 ng/ml of rhBMP-2. An apparent maximum change (about 2.6 times over the control,P<0.05) occurred at 6 hours. The second peak induction was about 2 times over the control and occurred at about 36 hours. After 48 hours of OP-1 treatment, the VEGF mRNA levels approached that of control.Conclusion RhBMP-2 can promote the expression of VEGF in a time-dependent manner. The VEGF mRNA level appears to be biphasic in rhBMP-2 treated cultures, showing peak induction at 6 and 36 h.
