rhIGF-Ⅰ、rhTGF-β1对骨髓基质细胞增殖分化成软骨细胞的实验研究
Effect of Recombinant Human Insulin-like Growth Factor-Ⅰ and Recombinant Human Transforming Growth Factor-β1 on the Proliferation and Differentiation of Bone Marrow Stromal Cells in Rabbits
【摘要】 目的研究重组人碱性成纤维细胞生长因子(rhbFGF-1)、重组人转化生长因子(rhTGF-β1)和重组人胰岛素样生长因子(rhIGF-Ⅰ)体外对骨髓基质细胞生物学活性的影响,并在体内诱导骨髓基质细胞向软骨细胞分化。方法单独或联合应用细胞因子对细胞进行体外诱导培养,进行常规染色、MTT、免疫组织化学染色观察细胞因子对骨髓基质细胞的作用。体内将rhTGF-β1和rhIGF-Ⅰ,骨髓基质细胞复合于纤维蛋白凝胶,植入到兔膝关节实验性关节软骨缺损,观察软骨修复效果。结果常规形态学观察,rhTGF-β1和rhIGF-Ⅰ联合应用诱导的细胞在形态上类似于软骨细胞,免疫组化染色提示诱导细胞具有软骨细胞表型。rhTGF-β1和rhIGF-Ⅰ在体内能诱导骨髓基质细胞向软骨细胞分化,修复缺损的软骨。结论rhT-GF-β1和rhIGF-Ⅰ联合应用可作为诱导骨髓基质细胞向软骨细胞分化的最佳组合。
【Abstract】 Objective To investigate the biological effect of recombinant human basic fibroblast growth factor-1 (rhbFGF-1),recombinant human transforming growth factor-β(rhTGF-β1) and recombinant human insulin-like growth factor-Ⅰ (rhIGF-Ⅰ) on the proliferation and differentiation of rabbit bone marrow stromal cells (bMSCs) in vitro, and on the differentiation from bone marrow stromal cells to cartilage cells in vivo.Methods The cytokines were used independently or in combination for inducing and culturing MSCs.The proliferation of cells was detected by MTT assay and the macroscopic histology. HE staining and immunohistochemical examinations were used in vivo to investigate the repair of the experimental articular cartilage defects in the knee joints of rabbits by using bMSCs combined with fibrin gel,rhTGF-β1 and rhIGF-Ⅰ.Results The cells induced by rhTGF-β1 and rhIGF-Ⅰ were similar to chondrocytes in morphology.The immunohistochemical examinations showed that the cells had the phenotype of chondrocytes.RhTGF-β1 and rhIGF-Ⅰ could induce the differentiaton from the bone marrow stromal cells into cartilage cells in vivo,and repair the articular cartilage defects. Conclusion The combination of rhTGF-β1 and rhIGF-Ⅰ as the best one can induce the differentiation from the bMSCs to cartilage cells.
