洛美利嗪对喹啉酸致海马神经元损伤的保护作用
Lomerizine protects the hippocampus neurons cultured in vitro from the injury caused by quinolinic acid
【摘要】 目的:利用洛美利嗪(LOM)研究钙离子拮抗剂对兴奋性氨基酸喹啉酸(QA)所致体外培养海马神经元损伤是否有保护作用。方法:原代海马神经元与QA共同培养建立兴奋毒损伤细胞模型,观察1.2×10-6m o l/L,1.2×1-0 7m o l/L,1.2×10-8m o l/L 3个剂量LOM对培养神经元的形态学、乳酸脱氢酶(LDH)、细胞存活率的影响。结果:LOM可提高QA损伤的海马神经元细胞存活率,减少LDH释放,与模型组比较差异有显著性(P<0.01),并能减轻神经元的病理形态学损伤。结论:LOM对QA所致的海马神经元损伤有明显的保护作用,而QA所引起的兴奋毒性可能与C a2+超载所造成的链式损伤有关。
【Abstract】 Objective:To study if Lomerizine(LOM) protects the hippocampus neurons cultured in vitro from the injury caused by quinolinic acid(QA).Methods:The model of neurons injured by QA was established by coculturing hippocampus neurons with QA.The morphological change,LDH and cell survival rate were examined after adding LOM into the medium with concentrations at 1.2×10-6mol/L,1.2×10-7mol/L,1.2×10-8mol/L.Results:LOM increased the survival rate of hippocampus neurons injured by QA,diminished the release of LDH and improved the pathological morphology,with significant difference from those of the control(P<0.01).Conclusion:LOM could remarkably protect the hippocampus cells from the damage of QA.
