Rh123、FDA荧光法测定超声作用对大肠杆菌细胞膜的影响
Determination of ultrasound-nduced alterations of Escherichia coli membrane with FDA and Rh123 fluorescent methods
【摘要】 目的研究大肠杆细胞经超声处理后细胞膜通透性和细胞膜电位的变化。方法应用二乙酸荧光素(FDA)、罗丹明123(Rh123)染色,通过荧光分光光度计检测FDA、Rh123荧光强度的变化,观察大肠杆菌细胞经25 kHz、300 W超声处理后细胞膜通透性和细胞膜电位的变化。结果大肠杆菌细胞经25 kHz 300W超声处理,随处理时间延长,FDA、Rh123都降低。在90 s内,超声处理对细胞存活率影响不明显,膜电位降低26.7%,膜通透性增加33.3%。结论大肠杆菌经一定超声处理后膜通透性增加,膜电位降低。
【Abstract】 In order to investigate the alteration of Escherichia coli(E.coli) membrane by ultrasound field,the cells were examined with fluorescence spectrophotometer after staining with Rh123,FDA at different time on ultrasound treatment.Alterations of fluorescence intensity were measured,which reflect sequential changes on E.coli membrane permeability,and membrane potential.The results indicated that Rh123 decreased gradually for ultrasound treating from 30 to 90 s,at 90 s decreased by 73.2% and the membrane potential decreased by 26.7%.FDA decreased gradually for ultrasound treating from 30 to 90 s at 300 W,at 90 s decreased by 66.7% and the membrane permeability increased by 33.3%.It was concluded that the membrane permeability of E.coli could be increased by the optimal ultrasound,that the membrane permeability and membrane potential could reflect the function of E.coli membrane and could be a simple and reliable assay for the change of membrane for ultrasound treating.
