【摘要】 目的探讨脑内注射凝血酶(thrombin,TM)对周围组织Caspase-3蛋白表达和细胞凋亡机制的影响。方法24只SD大鼠随机分为A,B,C3组,A组在大鼠右侧基底节区注入凝血酶(TM),B组注入TM+组织蛋白酶G(cathepsin G,CATG),分别于注射后6,24,48,72,96h时间点处死,C组注入生理盐水。取脑组织应用Western-blot技术检测Caspase-3蛋白表达。结果TM脑内注射后6h Caspase-3蛋白开始增加(P<0·01),24h达高峰(P<0·01),然后逐渐下降。TM+CATG脑内注射后Caspase-3蛋白表达与对照组比较差异均无显著性(P>0·05)。结论TM脑内注射后Caspase-3蛋白表达明显增加,TM通过蛋白酶激活受体-1激活(PAR-1)激活Caspase-3,诱导细胞凋亡。更多还原

【Abstract】 Objective To study caspase-3 protein expression and apoptosis mechanism after intracerebral injection of thrombin in rats.Methods 42 Sprague-Dawley rats were randomly divided into groups A,B and C. Thrombin, thrombin+cathepsin G or normal saline were stereotaxically injected into righ t caudate nucleus in rats of groups A,B or C respectively. Rats of group B were sacrificed at 6 h , 24 h , 48 h and 72 h . Caspase-3 protein expression was determined by western-blot. Results Caspase-3 protein expression increased at 6 h after intracerebral injection of thrombin(P<0.01), peaked at 24 h (P<0.01), and declined subsequently. Caspase-3 protein expression was not significantly different from control group after TM+CATG intracerebral injection (P>0.05). Conclusion Caspase-3 protein expression significantly increases after intracerebral injection of thronbin.Thrombin induces apoptosis by activating caspase-3 through PAR-1.更多还原
